The .45 Defender is a standard handgun that appears in Among Thieves, Drake's Deception, and Golden Abyss. It does not have a lot of ammo, and a below average rate of fire, but is generally accurate enough to serve well enough against soldiers equipped with the same weapon. Later on the 92FS - 9mm outperforms it entirely, but until then the .45 Defender is a reliable choice.
Chloe Frazer has a personal .45 Defender called Chloe's .45 Defender, which could be purchased in the Among Thieves store for $20,000, although Among Thieves Remastered changed the requirements to beating the game on Crushing.

Among Thieves[]
The Defender was first used by Nate, and Zoran Lazarević's soldiers in Borneo, as well as early on in the Nepalese city, until the weapon was replaced entirely with the more effective 92FS - 9mm. Chloe kept her customized .45 Defender however, which she was seen using in cutscenes in "The Dig" to shoot a soldier, in "Keep Moving" to hold Nate at gunpoint, and in "Tunnel Vision" to kill Draza.
Remastered[]
The .45 Defender is the only weapon that appears in Uncharted 3: Drake's Deception to not have it's HUD Icon updated to it's newer one. This was most likely an oversight and was not intentional.
Drake's Deception[]
In Drake's Deception, Nate once again used the Defender, but only for a limited time. Once he and Victor Sullivan traveled to Syria, he swapped it for a Para 9. The .45 Defender was a common firearm used by Hired thugs and early on in the chapters, Katherine Marlowe's agents. Chloe's .45 Defender is seen once when Nate, Cutter and Sully escape from the old North Atwood Station. When they open the gates to the streets, Chloe immediately shoots the agents behind them and then hides the gun. The gun can be seen in the chapter, "The Middle Way" when Talbot confronts Cutter, Nate, Chloe and Sully before she throws it off the edge.
The .45 Defender was also Salim's weapon of choice and is frequently seen firing it at Katherine Marlowe's agents in the final chapters of the game.
Multiplayer[]
The .45 Defender appeared in the multiplayer beta of Drake's Deception, however it was only available in the Co-Op game mode and appeared in the starting rounds by enemies. The final release of the multiplayer removed the gun from appearing at all which rendered it unused for the entire time that multiplayer was officially active even though you could get the gun through cheat commands.

The .45 Defender's sounds were enhanced in the Drake's Deception remaster for Uncharted: The Nathan Drake Collection.
Golden Abyss[]
In Golden Abyss, the gun has appeared rather sporadically during the first half of the game, serving as his starting weapon from the beginning.
